  1. Porcogen is a sterile solution of purified collagen extracted from Specific Pathogen Free (SPF) porcine skin. It contains stable collagen protein with a native triple-helical structure of molecular weight around 300 KD.

  3. Concentration:
    The concentration of Porcogen collagen is approximately 3.0 mg/ml . Small variations in concentration may exist from one production lot to another. The concentration for a specific lot will be labeled on the vial or bottle.
    pH:
    Porcogen collagen in dissolved in 0.01 N HCl, the pH of Porcogen collagen is approximately 2.0.
    Gel Time:
    Porcogen collagen can easily form collagen gel while the pH, concentration, ionic strength and temperature are at certain condition. The gel time may vary between lots because concentration and pH vary slightly between each lot.
    Purity:
    Porcogen is purified using GMP production process. It has Type I collagen specific banding pattern on SDS-PAGE result. This ultrapure (99.9%) collagen contains about 96-98% type I collagen with the remainder being Type III collagen.
    Endotoxin:
    Porcogen collagen contains <0.5 EU/ml endotoxin by Limulus Amoebocyte Lysate Assay.
    Lot Number and Expiration Date:
    The lot number and expiration date of each bottle of Porcogen has printed on the bottle label.

Storage and Handling

  1. Porcogen denatures under heat. It should be stored under refrigeration at 2-8 ℃ and used at temperature below 25 ℃ at pH 2 or below 40 ℃ while pH is raised to pH 7. Do not freeze.
    Aseptic handling is recommended to avoid contamination.

    Preparation of Porcogen collagen gel

1. Mix 9 parts of Porcogen collagen with 1 part of 10X phosphate buffer (0.2M Na2HPO4, 1.3M NaCl, pH = 7.4) or 10X cell culture media. Cells can be added to the mixed solution at this step or the next.
2. Adjust the pH to 7.2 ± 0.2 by titration with 0.1N NaOH or 0.1N HCl.
3. Warm up the mixed solution and keep temperature at 37 ℃. Gelation will form less than 45 min. For best results, allow at least 45 min. to an hour.

Packaging
Porcogen is available in 50 ml ,100 ml and 1L. Other concentrations and volumes are available upon request. It is suppluied in individual bottles and packaged in a polyethylene bag.
